Reconstitution of miR-3613-3p suppresses CDC7 expression and reduces the number of colony formation in A375-R cells. (A) Endogenous expression of CDC7 was evaluated in presence and absence of 2.5 µM Vemurafenib after 24 and 48 h by Western blot analysis in parental (A375-P & WM983B-P) and resistant (A375-R & WM983B-R) cells. (B) A375-R was transfected with 50 nM miR-3613-3p mimic for 36 h, RNA was extracted and qPCR was performed. (C) Cell lysate was collected from the transfected cells with miR-3613-3p mimic (25 and 50 nM) and Western blot analysis was carried out using anti-CDC7 and anti-p-ERK1/2 antibodies. (D) Transfected and scrambled cells were cultured in 6-well plates for colony formation assay and kept for 10 days. After staining, colonies were counted and expressed as a percentage to control cells. *Depicts significance at p < 0.05.
