At week 25 of the experiment, the control and HFD-fed mice were fasted for 14 h and administered insulin (1 U/kg body weight) intraperitoneally. Ten minutes later, the gastrocnemius muscle and whole skin, including both the dermis and epidermis, were isolated and western blotting was performed with anti-Akt, anti-phospho-Akt (p-Akt), anti-p70S6K, and anti-phospho-p70S6K (p-p70S6K) antibodies.
