PBMCs from healthy donors were pretreated with 100 μg/ml PGRN overnight and stimulated with K562 or 721.221 cells for 5 hours at an effector/target ratio of 10:1. The frequency of IFN-γ–producing and CD107a-expressing NK cells was determined by intracellular cytokine staining. (A and B) CD107a levels were measured in total NK cells (A) and CD56dim NK cells (B) by flow cytometry (n = 15).
