Liquid conjugation experiments in culture tubes with b pNuc-cis and c pNuc-trans were performed with 0.5 mm glass beads or without glass beads (filled diamonds) over 72 h at the indicated shaking speed (in revolutions per minute). Conjugations were performed with (filled circles) or without (filled diamonds) sgRNA targeting the STM1005 locus cloned into pNuc-cis and pNuc-trans. Both plasmids encoded the TevSpCas9 nuclease.
