Bone marrow cells, harvested from long bones of Notch3tm1.1Ecan mutant (black bars) and wild type littermate controls (white bars) were grown for 72 hours in the presence of M-CSF at 30 ng/ml and then seeded in the presence of M-CSF 30 ng/ml and RANKL at 10 ng/ml in the presence of anti-Notch3 NRR (N3NRR) or anti-ragweed control antibody (Ctrl) at 20 μg/ml for osteoclast formation. Representative images of TRAP-stained multinucleated cells are shown to the left. Data are expressed as total number of TRAP-positive multinucleated cells/well.
