Intracellular effect of 2′-OMe PGO on mycobacteria. (A) Confocal microscopy of RAW 264.7 macrophages infected with Mycobacterium smegmatis expressing mCherry Red fluorescent protein. The infected cells were treated with fluorescently labeled ald-PGO (FAM-PGO, 10 μM) for 24 h and analyzed by confocal microscopy. M. smegmatis is red, FAM-PGO is green, and nuclei stained with Hoecst33342 are blue. (B) Quantification of ald transcription in intracellular M. smegmatis. RAW 264.7 cells were infected with mycobacteria, treated with 20 μM of the indicated ASOs, and analyzed for ald mRNA expression by qRT-PCR.
