(A) CASIN prolongs lifespan of mice bearing bortezomib-resistant MM cells. NSG mice were subjected to myeloablative busulfan conditioning followed by intrafemoral injection of V10R cells. The mice were then treated with Vehicle, CASIN, bortezomib (BTZ), or CASIN + BTZ. Mouse survival was recorded. Data were analyzed using log-rank (Mantel-Cox) test, n = 10 mice. *P <0.05 vs. Vehicle, **P < 0.01 vs. Vehicle and #P < 0.01 vs. BTZ. (B) CASIN causes apoptosis of primary cells from bortezomib-resistant MM patients. Human CD138+ cells were isolated from peripheral blood of bortezomib (BTZ)-resistant MM patients. The cells were treated with or without CASIN (5 μM) for 2 days. Cell apoptosis was determined using flow cytometry analysis of Annexin V+ cells.
