A kindlin-3/leupaxin complex regulates paxillin tyrosine phosphorylation and podosome stability. Kindlin-3/leupaxin interaction is independent of integrin binding. However, kindlin-3 targets leupaxin into the podosome adhesion complex. In contrast, paxillin recruitment to podosomes occurs independent of kindlin-3. The recruitment of leupaxin into podosomes enables the tyrosine phosphatase PTP-PEST to dephosphorylate paxillin at Y31 and Y118, resulting in increased podosome lifetime and stability.
