Genetic inhibition of exemplary validated genes affects progranulin levels through transcriptional or posttranscriptional mechanisms in mouse cortical neurons. Mouse cortical neurons were transfected with Scr siRNA, Grn siRNA, Jmjd6 siRNA, Foxo1 siRNA, Trap1/Hsp90L siRNA, Tom1 siRNA, or Tsg101 siRNA, and cells were lysed for RNA extraction and qRT-PCR to measure Grn mRNA level, and conditioned media were used for ELISA to measure extracellular progranulin levels.
