Analysis of stress-inducible gene transcription in Synechocystis strains. Wild-type (WT), Δhik33 mutant, and ΔkatG/tpx double mutant strains were grown at 32 °C (control), then exposed to 22 °C for 30 min in the presence (+) or absence (−) of H2O2 at a final concentration of 0.25 mM. Control samples (32 °C) were also subjected to 0.25 mM H2O2. Transcription of Hik33-dependent (hliB, and sodB) and Hik33-independent (rbpA) genes was analyzed. Amounts of transcripts were assessed by qRT-PCR, normalized to those of the housekeeping gene rnpB, and expressed in % of the maximum value of gene expression in all samples [66].
