Ferrets (n = 6) were vaccinated once with a single dose as indicated followed by challenge with 106 TCID50 of wt A/CA07/2002 (H1N1). Two days after challenge (peak of viral replication) selected animals (n = 3) were euthanized to determine challenge virus loads in nasal turbinate (A) and lung (B) tissues. Nasal wash titers (C) were measured on days 0–5 post-challenge in the remaining 3 ferrets of each group. Dotted line indicates limit of detection. Nasal turbinate titers were not significantly reduced in FluMist® vaccinated ferrets at the highest dose of 1 x 107 PFU (p = 0063; Student’s unpaired t-test) compared to mock-vaccination. Undetectable titers are set at the limit of detection for the assay (1.5 log10). DPC: Days post-challenge.
