The effects of 23-mer PLP-antisense on (A) PLP and (B) MBP expression in OLs in primary cultures. Coverslips were incubated with 23-mer antisense to PLP, beginning on 13 DIV and continuing for 6 d (19 DIV). Coverslips were double-immunostained with A007 and PLP or MBP antibodies. The percentage of PLP+ OLs treated with antisense for 5 d remained below 10%, but the percentage of MBP+ OLs increased to 35%. The data for each time point were based on four different experiments; an average of three coverslips was counted for each group at each time point.
