Figure 5. Calcitriol-antigen liposomes induce antigen-specific PD-L1–dependent tolerance.

Figure 5

(A) BALB/c hosts received DO11.10 T cells and either anti–PD-L1 or isotype mAb. Naive or OVA/QuilA-primed mice (day 0) were administered calcitriol/OVA323–339 liposomes s.c. into the tail base on days 0 and 6. ILN or splenocytes were analyzed by flow cytometry on day 10 (naive) or 12 (primed). (B) Experimental conditions as for A in naive mice: MFI of PD-L1 in gated DiI+ and DiI dLN and spleen CD11b+Ly6C DCs 24 hours after the second liposome administration. (C) Experimental conditions were as in Figure 3G, except that groups of mice received either anti–PD-L1 or isotype mAb. n = 4–12 per group, representative of 2 replicates. *P < 0.05; **P < 0.01; ***P < 0.001; ****P < 0.0001, 2-way ANOVA with Tukey’s multiple comparison test.
