FCPR03 promotes translocation of Nrf-2 into the nucleus and increases the level of Nrf-2 in the cytoplasm after OGD. (A) HT-22 cells pretreated with FCPR03 (20 μM) for 1 h were subjected to 6 h of OGD. After fixing, cells were stained with Nrf-2. The intracellular localization of Nrf-2 was visualized by confocal microscopy. (B) The relative fluorescence intensity of Nrf-2 in the nucleus was quantified using image J (18 fields from three independent experiments, n = 3). (C, D, E, F) HT-22 cells pretreated with FCPR03 for 1 h were subjected to 6 h of OGD. The change of cytosolic and nuclear Nrf-2 protein levels were detected by Western blotting. The relative levels of Nrf-2/β-tubulin and Nrf-2/Histone H3 were measured by semiquantitative analysis of the Western blots (n = 5).
