A custom pathway map shows all reactions in the iSMU model. (A high-resolution image is available as Data Set S4 in the supplemental material.) S. mutans UA159 appears to lack complete pathways for synthesizing thiamine (A), riboflavin (B), pyridoxine (C), pantothenate (D), and biotin (E). Reactions are colored by agreement between the essentiality predictions of the associated genes and Tn-seq data from the work of Shields et al. (31). Blue reactions agree with the Tn-seq essentiality results; red reactions disagree. Overall agreement between the data sets is 84.8% (Fig. 4).
