Evaluation of disease resistance of T2 transgenic B. distachyon lines with Fg00677-RNAi, Fg08731-RNAi, and CYP51-RNAi constructs against F. graminearum. (A) The second spikelet from the top of the spike of transgenic B. distachyon lines and non-transgenic controls was point inoculated with F. graminearum PH-1 strain and photographed at 9 dpi. Bar, 1 cm. Arrows indicate the inoculation site. (B) Quantification of the disease symptoms using a scoring scale at 9 dpi. Values represent the means ± standard deviation of at least 30 plants. Similar results were obtained from three biological replicates. (C) F. graminearum and B. distachyon biomass ratio measured via total DNA content at 5 dpi by absolute quantification using the internal reference genes beta-tubulin and BdUBC18, respectively. Values represent the means ± standard deviation of three biological replicates. Differences were assessed using Student’s t tests. Double asterisks indicate P < 0.01.
