ICOS Agonist (Apt8a) Enhances T Lymphocyte Activation In Vitro

(A) Aptamer binding to cell detected by 32P-labeled aptamer incubated with different amounts of ICOS expressing lymphocytes or naive ICOS-null lymphocytes. (B) IFN-γ detected by ELISA in the supernatant of CD8 lymphocytes suboptimally activated with CD3 agonist antibody coated in 96-well plates and incubated with Apt8a aptamer or Apt ctrl. (Data are expressed as the mean ± SEM of triplicate cultures. The data are from one representative experiment out of two, with similar results.) (C and D) Proliferation measured by CFSE dilution of CD8 (C) and CD4 (D) isolated lymphocytes stimulated with CD3 agonist antibody and Atp8a or Apt ctrl. CFSE intensity was measured by flow cytometry on the CD8 or CD4 gated population (The data are from one representative experiment out of two with similar results.)
