The observed heat capacity profile of a 12 bp all-CG duplex. The expected heat capacity of the fully folded DNA duplex is obtained by subtracting the heat capacity increment, ΔCp, (12 × 0.13 = 1.56 kJ K−1 mol−1), from the heat capacity of DNA at 110 °C and linearly extrapolating back to the start of melting at 0 °C. The experimental excess heat effect is then deconvoluted into non-cooperative (gradual, vertical hatching) and cooperative (horizontal hatching) phases (Vaitiekunas et al. 2015)
