Reovirus activates STAT1 and STAT2 signaling, and topoisomerase inhibitors activate DNA damage response pathways. (A) MDA-MB-231 cells were treated with the vehicle (DMSO) or 2 μM doxorubicin, epirubicin, or topotecan for 1 h; infected with reovirus at an MOI of 100 PFU/cell; and incubated with DMSO or 1 μM topoisomerase inhibitors for 0 to 2 days postinfection. Whole-cell lysates were resolved by SDS-PAGE and immunoblotted with antibodies specific for phosphorylated and total STAT1, STAT2, STAT3, ATM, p53, and GAPDH and reovirus. Residues recognized by phosphorylation-specific antibodies are shown in parentheses. (B and C) Quantitative densitometry of immunoblots from three independent experiments.
