MK-2206 and WZB117 affect proteins involved in DNA damage signaling and repair, determined by Western blot analysis, and induce DNA damage, determined by the comet assay. (A) MCF-7 cells. (B) MDA-MB-231 cells. MCF-7 cells were treated with 6 µM MK-2206 and 60 µM WZB117 and MDA-MB-231 cells were treated with 12 µM MK-2206 and 60 µM WZB117, either alone or in combination, for 24 or 48 h. Cells were then harvested for Western blot analysis. Quantitative data are presented as the mean ± SEM of three independent experiments. (C) Comet images. (D) Quantitative data of comet assay.
