Ser10 phosphorylation of P27^Kip1 mediates the effect of CACYBP on HCC cells. (A) Western blotting analysis of cell cycle proteins after reconstitution with P27^Kip1 phosphomimetic mutants in CACYBP-depleted SK-Hep-1 cells. Cells were transfected with the indicated plasmids and incubated for 24 h. Cell lysates were subjected to immunoblotting analysis with the indicated antibodies. (B) P27^Kip1-S10D overexpression rescued the apoptotic phenotype after CACYBP depletion in SK-Hep-1 cells. Cells were transfected with the indicated plasmids and incubated for 24 h.
