(A) MOLM-13 and MV4–11 cells were treated with midostaurin and venetoclax for 4 hours. Total RNA was isolated and Mcl-1 transcripts were determined by real-time RT-PCR using TaqMan probes. *indicates p<0.05, **indicates p<0.01, and ***indicates p<0.001. (B-C) MOLM-13 and MV4–11 cells were treated with midostaurin and/or venetoclax for 4 h, washed, and then treated with cycloheximide for up to 2 hours. Whole cell lysates were subjected to Western blotting. The fold changes for the Mcl-1 densitometry measurements, normalized to β-actin and then compared with no drug treatment control, are shown.
