Cytotoxic effects of PM2.5 in human peripheral blood mononuclear cells (PBMC). (A) PBMC from six study subjects were exposed to PM2.5 at final concentrations of 0, 1, and 5 µg/mL for 0, 1, 4, and 7 days. As a measure of cytotoxicity, leakage of lactate dehydrogenase (LDH) into culture supernatants was determined. (B) Cytotoxic effects of PM2.5 on PBMC from ten study subjects pre-exposed to PM2.5 for 20 h followed by infection with M.tb at multiplicities of infection (MOI) 1 for 0 (2 hours), 1, 4, and 7 days were determined by measuring LDH concentrations in culture supernatants. Percent LDH expression is expressed as 5/95 percentile box-and-whiskers where the center represents the 50th percentile, the upper hinge the 75th percentile, and the lower hinge the 25th percentile.
