Biphasic regulation of cAMP production persists after knockdown of Gs. HEK-TSHR cells were transfected with human GNAS (Gs) siRNA. Control cells were transfected with non-targeting pool siRNA. Seventy-two hours after transfection, the cells were stimulated with increasing doses of TSH for 1 hour. Subsequently, the cells were lysed, and cAMP production was determined. The data represent the mean ± S.E.M. of three experiments in duplicate. KD, Knockdown.
