Figure 1. — IKKβ phosphorylates NF-κB p50. (A) HEK293T cells were transfected with FLAG-IKKβ^K44M (kinase dead) or FLAG-IKKβ^SSEE (constitutively active) as indicated. IKKβ was immunoprecipitated from whole cell lysates with anti-FLAG antibody and incorporated in a kinase assay using recombinant GST-p50 or recombinant GST. (B) In vitro kinase assay employing recombinant IKKβ and recombinant GST-p50 or GST. (C) HEK293T cells were co-transfected with Myc-p50 and FLAG-IKKβ^SSEE or FLAG-IKKβ^K44M as indicated. p50 was immunoprecipitated (IP) from whole cell lysates with anti-Myc antibody and analysed by western blot (WB) using the indicated antibodies.
