LGS inhibits the STAT3 activation in LPS-induced RAW264.7 cells. (A−C) LGS inhibits the STAT3 signaling pathway. RAW264.7 cells were cultured with various concentrations of LGS in the presence or absence of LPS (1 μg/ml). The protein expressions of P-STAT3 (Tyr705) and STAT3 were measured by Western blotting (A) and confocal microscopy (600 ×) (B and C), respectively. ### P < 0.001 versus control, *P < 0.05, **P < 0.01, ***P < 0.001 versus LPS treatment by one-way ANOVA with Tukey’s test. (D) LGS suppresses the transcriptional activity of STAT3. The RAW264.7 cells were incubated in 96-well plates for 24 h. After infected by adenovirus, cells were treated with LGS (200 μg/ml) for another 24 h. The luciferase activity was detected by Dual-Luciferase® Reporter Assay System.
