Schematic representation of the process leading to the inclusion of an aqueous solution containing a biomolecule in its active form in PDMS. After pouring a thin basal layer of liquid PDMS that is then cured, a subsequent thicker layer is added on top. Then the biomolecule-rich solution is deposited in µl-sized droplets and the PDMS is let to cure under mild condition, to be characterized subsequently by in situ spectrophotometry or ex situ using biochemical methods.
