Genetic stability of rSA11/NSP3-FL-UnaG. Virus was serially passaged 10 times (P1 to P10) in MA104 cells, using 1:1,000 dilutions of infected cell lysate as the inoculum. RNA was recovered from infected cell lysates using TRIzol, treated for 30 min with RNase T1 at room temperature, and resolved by electrophoresis on an 8% polyacrylamide gel. RNA was detected by staining the gel with ethidium bromide. The red arrowheads denote the position of the segment 7 (NSP3) RNA. wt, wild-type SA11 rotavirus.
