PPI suppressed lncRNA HOTAIR levels and increased c-Jun protein expression and silencing of c-Jun neutralized the PPI-inhibited HOTAIR expression. (A) PC9 and H1650 cells were treated with different concentrations of PPI for 24 h. The expression of c-Jun protein was detected by Western blot. GAPDH was used as a loading control. (B) PC9 and H1650 cells were treated with PPI (1.6 μM) for 24 h. The expression of HOTAIR was detected by qRT-PCR. (C) Cellular protein was isolated from PC9 and H1650 cells transfected with the control or HOTAIR expression vectors for up to 24 h before exposing the cells to PPI (1.6 μM) for an additional 24 h. Afterward, the expressions of c-Jun proteins were detected by Western blot. GAPDH was used as a loading control.
