Figure 3.

Vgl1 regulates H3K9 methylation and Swi6 recruitment to pericentromeric heterochromatin. A–D, ChIP experiments showing that loss of Vgl1 causes decrease in H3K9me2 and Swi6 binding at the centromeric dg and dh repeats. E and F, H3K9me2 levels and Swi6 recruitment was also dramatically reduced at the imr1::ura4 reporter gene in Vgl1-deleted cells. G, Western blotting of Swi6 protein levels in WT and vgl1Δ cells. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as a loading control. H, ChIP analysis showing increased H3K14Ac levels upon Vgl deletion at dg repeats. Clr4Δ and Swi6Δ strains were used as a control in all the ChIP experiment, and error bars represent standard deviations from three independent experiments.
