Gating strategy to identify differentiation stages of T-cells. Lymphocytes were selected with a SSC/CD45 gate. Then, CD4+ and CD8+ T-cells were identified as CD3+CD8- or CD3+CD8+ lymphocytes, respectively (upper left). CCR7 expression on CD4+ and CD8+ T-cells is shown in a histogram (upper right). Finally, T-cell maturation stages were defined based on the differential expression of CCR7, CD45RA, and CD45RO on CD8+ (lower left) or CD4+ T-cells (lower right). TN = naïve T-cells (CD45RA+CCR7+); TCM = central memory T-cells (CD45RO+CCR7+); TEM = effector memory T-cells; TEMRA = CD45RA+ effector memory (CD45RA+CCR7-).
