Effects of PKM2 knockdown on migration and invasion of 786-O cells. (A) A wound was formed and migration of 786-O cells was measured every day using a phase-contrast microscope for 72 h. (E) Cells were harvested and whole cell lysates were examined by Western blotting using antibodies specific for matrix metalloproteinase (MMP)-2, MMP-9, N-cadherin, E-cadherin, and vimentin. GAPDH was used as internal loading control. (F) Band intensities were measured and plotted as a bar graph relative to GAPDH level. One-way ANOVA was used to compare means of different groups. Differences between means were considered significant at p < 0.05 using Tukey’s multiple comparison test; *** p < 0.001 as compared with normal control cells.
