RoGalU2 enzyme activity depending on the glucose 1-phosphate concentration between 50 µM and 500 mM at a fixed UTP concentration of 1.5 mM. Data for glucose 1-phosphate concentrations between 0 and 10 mM were fit to the kinetic model of Michaelis–Menten (a). The data obtained over the total glucose 1-phosphate variation are presented in (b), whereas no fit was able to describe the data. Assay mixture contained 0.05–500 mM glucose 1-phosphate, 1.5 mM UTP, 1 mM MgCl₂, 50 mM Hepes buffer, pH 7.4, 1.7 µg RoGalU2. Reaction was carried out at 42 °C in 1 mL volume. Means with standard deviations are shown from a minimum of triplicates.
