Role of AC8 in focal adhesion kinase (FAK) tyrosine phosphorylation in MDA-MB-231 cells. MDA-MB-231 cells were transfected with siAC8#1, siAC8#2, or scramble plasmid (Sc), as indicated. Forty-eight hours later, cells were lysed and subjected to 10% SDS-PAGE and Western blotting with anti-phospho-FAK (Y³⁹⁷) or anti-FAK specific antibodies. Membranes were reprobed with anti-β-actin antibody for protein loading control. Blots are representative of five separate experiments. The box plot represents FAK tyrosine phosphorylation presented as the phospho-FAK/total FAK ratio.
