Structure, physicochemical properties and cytotoxic potential of AS2717638 and compound 3. Chemical structures of AS2717638 (A) and compound 3 (C). SwissADME was used to calculate physicochemical properties relevant to the “rule of five” that predict drug-likeliness of a compound (Lipinski et al., 2001). Cytotoxicity was assessed using the MTT assay. BV-2 microglia cells were cultured in 6-well plates, serum-starved overnight (o/n) and incubated with the indicated concentrations of AS2717638 (B) and compound 3 (D) in DMSO for 2 and 24 h. DMSO was used as vehicle control. For convenience, the names of the inhibitors are presented as AS and comp3. Results from three independent experiments in triplicates are presented as mean ± SD (##p < 0.01, ###p < 0.001 compared to vehicle control; one-way ANOVA with Bonferroni correction).
