In vitro validation of the KdsD-FsaA pathway for GA production. In (A) In vitro enzyme assay scheme for conversion of D-ribulose 5-phosphate into GA. In (B) is shown the NADH production resulting from the conversion of D-ribulose 5-phosphate into GA. The reaction was made in 1 ml buffer solution (100 mM Tris-HCl pH 7.5, 10 mM Mgcl2, 0.1 mM MnCL2, and 1 mM sodium thiamine pyrophosphate) in the presence of 5 mM D-ribulose 5-phosphate, 2 mM NAD+, and 10 μg of each of purified KdsD, FsaA, and AldA enzyme.
