The following pH buffer systems were used: 50 mM sodium acetate buffer (pH 3.0–5.0), 50 mM Na2HPO4 (pH 6.0–7.0), 50 mM Tris-HCl buffer (pH 8.0–9.0), and 100 mM NaHCO3 (pH 10.0–11.0). (B) The effect of different temperatures on the enzyme activity at pH 6.0. (C) Stability and activity of the enzyme under different pH buffers. Dep42 was preincubated in different pH buffers for 30 min, and the residual activity was measured at 25°C. (D) Effect of temperature on enzyme stability and activity. Dep42 was preincubated at different temperatures for 30 min, and the residual activity was measured at pH 6.0. Relative enzyme activity was presented as a percentage of the highest activity value.
