Candidate gene pool induces S-phase re-entry in cultured adult mouse cardiomyocytes. (a) Lineage-traced cardiomyocytes transduced with a pool of candidate genes show S-phase activity via EdU labeling of in situ fixed cardiomyocytes. (b) Quantification of S-phase induction by pooled candidate genes compared to known cardiomyocyte cell cycle regulators p38i/FGF-1 and constitutively active Yap (caYap). Ad-GFP was used as a control for Adenovirus treatment. Negative control is no virus. (c) A subpool comprising genes 1–17 contains a candidate gene that is capable of activation of EdU incorporation in adult cardiomyocytes. (d) Identification of E2F2 that is sufficient to induce adult cardiomyocyte S-phase entry. Error bars represent standard deviation of mean from greater than or equal to 3 independent experiments.
