PRKD2 promoted proliferation and drug resistance of AML cells by regulating apoptosis. (A) The mRNA expression levels of genes in K562 and K562-A02 (drug-resistant cell line) were detected by qRT-PCR. (B, C) KASUMI and U937 cells were infected with PRKD2 lentivirus and negative control (NC). The mRNA level of PRKD2 was detected by qRT-PCR. The protein level of PRKD2 was detected by Western blot. (D) CCK8 assay showed that overexpression of PRKD2-promoted proliferation of AML cells. (E) Cells were incubated with ADM for 48 hrs and cell viability was detected by CCK8 assay. The results showed overexpression of PRKD2 decreased the sensitivity of AML cells to ADM. (F) Cells are treated with 0.4 μg/mL (KASUMI) or 0.3 μg/mL (U937) ADM for 48 hrs, and flow cytometry showed overexpression of PRKD2 inhibited ADM induced apoptosis of AML cells. *P < 0.05, **P < 0.01, ***P < 0.001.
