Knockdown of PRKD2-inhibited drug resistance by inducing apoptosis. (A, B) PRKD2 expression was knocked down by siRNAs in KASUMI and U937 cells. The mRNA level of PRKD2 was detected by qRT-PCR. The protein level of PRKD2 was detected by Western blot. (C) KASUMI and U937 cells were treated with ADM for 48 hrs, and cell viability was detected by CCK8 assay. The results showed knockdown of PRKD2 increased the sensitivity of AML cells to ADM. (D) Cells were treated with 0.4 μg/mL (KASUMI) and 0.3 μg/mL (U937) ADM, and flow cytometry showed knockdown of PRKD2 increased ADM induced apoptosis of AML cells. *P < 0.05, **P < 0.01, ***P < 0.001.
