Oxymatrine induces the apoptosis of MCF-7 cells. (A) MCF-7 cells were pre-incubated with the indicated concentrations of oxymatrine for 24 hrs, and then cells were stained with Hoechst 33258 and observed with a fluorescence microscope. (B) MCF-7 cells were pre-incubated with the indicated concentrations of oxymatrine for 24 hrs, and cells were treated with ANNEXIN-V-FITC and analyzed by FACS analysis. The experiment was repeated thrice. (C) MCF-7 cells were treated with DMSO or the indicated concentrations of oxymatrine for 24 hrs, and expression of PI3K, AKT, Akt^p-Thr308 caspase-3, Bax and Bcl-2 was measured by Western blotting. (D) MCF-7 cells were treated with DMSO or the indicated concentrations of oxymatrine for 24 hrs, and mRNA expression of caspase-3, Bax and Bcl-2 was measured by real-time RT-PCR. Results are the mean ± SD of three experiments done in triplicate. **P < 0.01, compared with the control group.
