(A) Cleavage of proISG15 substrates derived from different animal species by nairovirus OTUs. Each OTU was present at a 20 nM concentration and run against 10 μM of each ISG15 at 37°C. Samples were taken at the indicated timepoints and the reaction quenched in 2x Laemmli sample buffer followed by boiling at 98°C for five minutes. SDS-PAGE analysis was performed using Mini-PROTEAN® TGX Stain-Free as described in the Materials and Methods. The substrate preference of OTUs was assessed by comparing the relative quantities of cleavage product versus unreacted substrate over the time course of the assay.
