Actin polymerization at the LS was analyzed by confocal microscopy of untreated (A) WT and (B) Tlr4−/− BMMs, or (C-D) pre-treated with TLR4 agonist MPLA (1 μg/mL) for 1 hr, prior to incubation with A546-agLDL for 1 hr in presence of agonist. (E) Myd88−/− and (F) Cd14−/− BMMs were also incubated with A546-agLDL for 1 hr. Cells were fixed, and F-actin stained using A488-phalloidin. Arrows highlight areas of macrophage actin polymerization at sites of contact with agLDL. (G) Confocal images were used to quantify F-actin colocalized with agLDL per field. Data were compiled from at least 3 independent experiments per condition. Error bars represent the standard error of the mean (SEM). *** p ≤ 0.001. n.s. not statistically significant (p > 0.05).
