Evaluation of lysis buffer shelf life. Human keratinocytes (human adult low calcium high temperature cells, HaCaTs) were irradiated with ultraviolet B radiation (UVB), lysed, and electrophoresed following the standard protocol. The cells were then (A) lysed or (B) neutralized in neutralization buffer, each of which were made fresh on the day of the experiment (0 weeks), 1, 2, 4, or 8 weeks before the experiment, and stored at room temperature. Tukey box and whisker plot of median and inter-quartile range for results from two individual experiments, 100 determinations per treatment per experiment, and analyzed by Kruskal–Wallis test with Dunn’s multiple comparisons test. ns = not significant. Also shown are representative comet images of control and cells irradiated with 1 J/cm2 UVB (inset images).
