32 mg/mL PEGVS constructs were incubated with the broad-spectrum MMP inhibitor GM6001 for 7 d. The shear storage modulus (G’) at 7 d was measured in each condition (A, *: p < 0.05, relative to vehicle 7 d). There was a modest trend toward lower cell density with increasing GM6001 concentration but did not reach statistical significance (B, ANOVA: p = 0.0734). Average cell circularity, a measure of cell spreading, increased with GM6001 concentration (C, #: p < 0.01, relative to 10 μM GM6001; &: p < 0.05, relative to 1 μM GM6001, lower values of circularity reflect greater irregularly shaped spreading). Projected cell area covered also decreased with GM6001 concentration but did not reach statistical significance (D, ANOVA: p = 0.1417). N=3.
