ERα plays a critical role in E2-dependent reporter gene activity in Ishikawa cells expressing ERβ5. Overexpression of ERβ5 in ERα^pos Ishikawa cells (ratio mRNAs ERβ5: ERα = 1.5:1) resulted in a significant increase in reporter gene compared to cells treated with vehicle (24 h ± E2). Targeted knockdown of ERα abrogated response to E2. Results are displayed as fold difference compared to vehicle: triplicate experiments performed in triplicate wells. Statistical analysis was performed by one-way ANOVA with Tukey’s post hoc test, *P < 0.05, **P < 0.01, ***P < 0.001, ****P < 0.0001.
