BPTF knockdown inhibited COX-2 promoter activity and COX-2 expression. A. A549 cells were co-transfected with BPTF siRNA and COX-2 promoter driven luciferase plasmid for 48 hours. Then the luciferase expression was analyzed by a microplate reader (***P<0.001). B. NCI-H460 cells were also co-transfected with BPTF siRNA and COX-2 promoter driven luciferase plasmid, and then analyzed by the microplate reader (*P<0.1). C. A549 and NCI-H460 cells were transfected with BPTF specific siRNA and the mRNA expression of BPTF and COX-2 was analyzed by RT-PCR. D. BPTF was knocked down by BPTF siRNA, and the protein expression of BPTF and COX-2 was detected by Western blot in A549 and NCI-H460 cells. E. Ten nude mice were injected with A549 cells and tumors grew to 5 mm×5 mm (length × width) after 2 weeks. Nude mice were randomly divided into 2 groups. BPTF shRNA and control shRNA were respectively injected into the tumors once every 4 days. After 28 days, the mice were sacrificed to examine the expression of BPTF and COX-2. Representative results from two pairs of nude mice were shown. F. Image Lab software was used to analyze the protein bands of BPTF and COX-2 in the two groups of nude mice.
