mTOR and SREBP activity is required for TLR4-dependent upregulation of fatty acid synthesis. C57BL/6J BMDM were treated with the vehicle or 100 ng/ml KLA for 8 h and then treated for an additional 8 h with 250 nM Torin1 or 10 μM PF-429242 in the continued presence of KLA. Following treatment, mRNA levels of Srebf1c (A), Fads2 (B), Scd2 (C), and Lxrα (D) were quantified by real-time PCR and normalized to the value for cyclophilin. *, statistically significant difference between the vehicle and KLA within each pair determined by 2-way ANOVA (P ≤ 0.05; n = 8).
