Plasma membrane vesicles made from the S837A strain have more Pdr5 in the membrane. Purified PM vesicles were prepared as described by Kolaczkowski et al. (1996) and modified by Ernst et al. (2008). (A) PM (6 μg) protein samples from three preparations of S837A (JG2175) and three preparations of WT (JG2015) PM vesicles were solubilized in SDS PAGE and subjected to electrophoresis in 7% tris-acetate gels for 80 min at 150 V before staining with coomaissie blue. (B) Samples (3 μg) from the same PM vesicle preparations were also immunoblotted as described in the Materials and Methods.
