a ZT-1a dose dependently inhibited KCC3 Thr991/Thr1048 phosphorylation in cells. HEK-293 cells were transfected with a DNA construct encoding wild-type N-terminally FLAG-tagged KCC3. Cells at 36 h post transfection were exposed for 30 min to either control isotonic or hypotonic low [Cl] conditions, and then treated in the same conditions for an additional 30 min in the presence of inhibitors at the indicated concentrations. Cell lysates were subjected to SDS-PAGE and immunoblot with the indicated antibodies. b Immunoblot quantitation on ZT-1a, Closantel, STOCK1S-14279, and STOCK1S-50699.
